基于网络药理学研究黄精丸防治阿尔茨海默病的作用机制

[摘要]目的采用网络药理学的技术和方法,探讨黄精丸治疗阿尔茨海默病(AD)的作用机制。方法通过中药系统药理数据库及分析平台(TCMSP)筛选黄精丸的潜在活性成分,DrugBank数据库预测化合物靶点;GeneCards和OMIM数据库收集疾病靶点,构建韦恩图,得到黄精丸治疗AD的作用靶点,通过Cytoscape 3.8.1 软件生成药物-成分-靶点网络;R 4.0.3软件进行基因本体(GO)功能注释和东京基因组百科全书(KEGG)通路富集分析。结果共筛选出黄精丸活性成分12个,黄精丸与AD疾病共同靶点92个,重要活性成分包括黄苓素(baicalein)、5,4’-二羟基黄酮(4’,5-Dihydroxyflavone)、苷草素(DFV)等;核心靶点包括AKT1、TP53、VEGFA、SIRT1、EGFR、PIK3CA、PIK3CB、NOS2、MAPK3等;KEGG 主要通路富集在癌症信号通路,代谢通路,PI3K-Akt信号通路、神经退行性性变信号通路、AD等。结论黄精丸可能主要通过多成分、多靶点、多通路,发挥对AD的治疗作用。

Investigation on the action mechanism of HJ pill in the prevention and treatment of Alzheimer’s disease based on network pharmacology

[Abstract] Objective To investigate the action mechanism of HJ pill in the treatment of Alzheimer's disease (AD)based on the technology and method of network pharmacology. Methods The potential active components of HJ pill were screened by traditional Chinese medicine systems pharmacology (TCMSP),and the DrugBank database was used to predict the compound targets. Disease targets were collected with GeneCards and OMIM databases to construct a Wayne map. The action target of HJ pill in the treatment of AD was obtained,and the drug-component-target network was generated by Cytoscape 3.8.1 software. R 4.0.3 software was used for gene ontology (GO)functional annotation and Tokyo Genome Encyclopedia (KEGG)pathway enrichment analysis. Results A total of 12 active components of HJ pill and 92 common targets of HJ pill and AD were screened. Baicalein,4' ,5-Dihydroxyflavone,glycyrrhizin (DFV),etc. were included in the important active components. AKT1,TP53,VEGFA,SIRT1,EGFR,PIK3CA,PIK3CB,NOS2,MAPK3,etc. were included in the core targets. The main pathways of KEGG were concentrated in cancer signal pathway,metabolic pathway,PI3K-Akt signal pathway,neurodegenerative signal pathway,AD and so on. Conclusion HJ pill may play a therapeutic role in AD mainly through multiple components,multiple targets and multiple pathways.
