基于数据挖掘探究兰艾双香方诱导大肠杆菌K12菌株铁死亡机制研究

[摘要]目的本研究旨在通^数据挖掘探究兰艾双香方在发挥抑菌作用过程中,常见致病菌―大肠杆菌诱使铁死亡发生的过程,为后续开展常见致病菌发生铁死亡的研究提供基础研究方向和理论基础。方法以兰艾双香方中的各种中药在中国知网、万方数据、维普以及中药系统药理学数据库等检索有效成分及对应蛋白。建立中药数据的 Excel 表格,对有效成分、分子蛋白等进行分析,并分析药物蛋白在大肠杆菌 K12菌株不同样本起作用蛋白,最后对在大肠杆菌 K12菌株中铁死亡蛋白进行分析。结果兰艾双香方中共138个活性化学成分,共获得兰艾双香方蛋白265个,其中“MG1655”样品中共有17个作用蛋白,“W3110”样本中共有5个作用蛋白,fhuA是在大肠杆菌 K12抑菌过程中诱导大肠杆菌发生铁死亡的关键蛋白。结论通过数据发掘的方式,发现兰艾双香方中存在的铁色素外膜转运蛋白在大肠杆菌铁死亡进程中发挥作用,为后续开展诱导常见大肠杆菌等常见致病菌发生铁死亡从而发挥抑菌作用提供了基础。

Investigation on ferroptosis mechanism of Escherichia coli K12 strain induced by Lan'aiShuangxiang Formula based on data mining

[Abstract] Objective To investigate the process of ferroptosis of common pathogenic bacteria Escherichia coli in the process of bacteriostasis effect of Lan'aiShuangxiang Formula based on data mining, so as to provide basic research direction and theoretical basis for the follow-up research on ferroptosis of common pathogenic bacteria. Methods The effective components and corresponding proteins were retrieved in CNKI, Wanfang data, VIP and TCMSP with Lan'aiShuangxiang Formula. Excel table of Chinese materia medica data was established to analyze the effective components and molecular proteins. And the effective proteins of drug proteins in different samples of Escherichia coli K12 strain were analyzed. Finally, the ferroptosis protein in Escherichia coli K12 strain was analyzed. Results A total of 138 effective chemical components were found in the Lan'aiShuangxiang Formula, and 265 Lan'aiShuangxiang Formula proteins were obtained. In these proteins, there were 17 effective proteins in “MG1655”sample and 5 effective proteins in "W3110" sample. fhuA was the key protein to induce the ferroptosis of Escherichia coli in the process of bacteriostasis of Escherichia coli K12. Conclusion Based on data mining, it was found that the iron pigment outer membrane transporter in Lan'aiShuangxiang Formula played a role in the process of ferroptosis of Escherichia coli. It provides basic research for the subsequent induction of common pathogenic bacteria such as common Escherichia coli to play a bacteriostasis role through ferroptosis.
